The MicroCal VP-ITC (Isothermal Titration Calorimeter) unit directly measures heat evolved or absorbed in liquid samples as a result of mixing precise amounts of reactants. A spinning syringe is utilized for injecting and mixing of reactants. The user inputs the experimental parameters (temperature, number of injections, injection volumes) and the computer carries out the experiment. Origin® software is then used to analyze the ITC data using fitting models to calculate reaction stoichiometry (n), binding constant (KD), enthalpy (ΔH) and entropy (ΔS).
To avoid damage or contamination, researchers must provide their own Hamilton 81414 syringe for loading sample solutions. The syringe can be purchased through VWR via CyBuy or directly from Hamilton. The instrument must be reserved in advance, and various rules apply. Reserve time using iLab. Reservations do not require approval.

The instrument is to be used by students and other researchers. Significant training is required, including sucessful completion of several tutorials and lessons before you will be authorized to use the instrument. Staff will NOT provide methods development assistance for this instrument, but will direct you to internet-based ITC resources. The hourly rates are listed below. NOTE: There is a minimum charge of $10 for each use of the instrument.

Product Overview
MicroCal PEAQ ITC Automated combines the high sensitivity of the MicroCal PEAQ ITC with walkaway automation to meet the productivity needs of busy research and drug discovery laboratories. Temperature controlled 4 x 96 well plates and user guided software together enable exceptional sensitivity and high quality data with robust operation. ITC is an essential tool in drug discovery and enables the study of biomolecular interactions as well as changes in conformation such as protein unfolding. It can also be used to measure enzyme kinetics.
Microcal Peaq-itc Analysis Software
MicroCal PEAQ ITC Automated measures heat released or absorbed during biochemical binding events to calculate binding affinity (K D ), stoichiometry (n), enthalpy (ΔH) and entropy (ΔS) in a single experiment. The system uses Isothermal Titration calorimetry (ITC), a technique that requires NO labelling and minimal assay development.
The PEAQ ITC micro calorimeter comprises two cells — a sample cell in which the titration experiment is performed, and a reference cell which contains only water. A series of small aliquots of ligand are injected into the sample or protein solution. If binding occurs between the ligand and protein, heat changes of a few millionths of a degree Celsius are detected and measured until the binding reaction has reached equilibrium.
